Mass Spectrometry

Quillen College of Medicine

Our Equipment

Courtesy of ThermoFisher

Our Proteomics and Mass Spectrometry Facility is equipped with a state-of-the-art ProteomeX Station (ThermoFisher), which includes a Finnigan LTQ XL Linear Ion Trap Mass Spectrometer (LITMS) and integrated LC system with auto-sampler. For more details see LTQ XL specifications.

Linear Ion Trap technology greatly increase the sensitivity of MS analyses and allows up to 12 consequent MS/MS scans (MS n). Plea.

NSI Head with a nano-column

The LTQ XL allows electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI); however, the unique feature of this device is nanospray ionization (NSI), which allows detection of peptides and chemicals in the femtomole range. We use NSI on an every-day-basis both in dynamic mode (nano-column injection, LC/MS analysis) and in static mode (direct injection from a nano-tip, MS analysis).

The LTQ XL is capable of analyzing complex protein samples ("full digests" of complex mixtures of proteins or peptides), as well as the identification of posttranslational protein modifications. See these examples of complex peptide mixture analysis, as well as identification of glycosylated peptides, and phosphorylated peptides with MS 3 data-dependent analysis (courtesy of ThermoFisher).
